Enterohemorrhagic Escherichia coli (EHEC) are an important cause of bloody diarrhea and hemolytic uremic syndrome (HUS. In cooperation with hospitals in the Brussels Capital-Region, human stool samples willbe collected and analyzed for EHEC in order to investigate the incidence of this food pathogen. Cases of HUS will be registered in a surveillance program, and patient's samples will be analyzed using highly sensitive techniques. Patients who are found positive for EHEC will be enrolled in a case-control study to investigate the relationship between nutritional habits and the occurence of EHEC and HUS. Also, this will allow us to identify important sources of infection and routes of transmission. where possible, samples of putative sources of infection will be collected and processed for analysis. During this project, we will optimize protocols for new techniques, such as PCR-RFLP, PHIA and real-time PCR, and evaluate their use in the clinical laboratory. New methods for EHEC subtyping, such as sequence analysis, multiplec PCR, and MLVA, will allow us to faster identify communal spreading of EHEC strains and prevent epidemic outbreaks. using the information collected in the case-control study, measures will be proposed to prevent human infections and published in a brichure to inform the public about this emerging pathogen.
